Sen was born as Shubhomitra Sen in Kolkata, West Bengal. Rimi completed her schooling from Bidya Bharati Girls' High School in the year 1998.
In 2006 Sen made racist comments about black African people in an online interview about a film, saying, "I play a sweet and beautiful girl in the film.

Her younger sister Shamita Shetty is also a Bollywood film actress. Shilpa has also judged a reality show "Zara Nachke Dikha" along with two other judges Arshad Warsi and Vaibhavi Merchant aired in Starplus.
